1977 Johnson 115 outboard no water flow

This far we have replaced the impeller, ensured it is locked onto the drive shaft securely, and run air though all the water holding lines we can. We have removed the thermostat and found no blockages... it no water circulating. Does the pump need to be primed? We are running it in a horse trough so the intakes are well below water level. Need a few ideas...

we can put the motor in drive and reverse so it appears the drive shaft is turning.

Your horse trough isn't deep enough. The water level must be higher than the intakes and should be well above the parting line between the lower unit and mid-section.

The impeller must be submerged, it doesn't suck water. It moves water by being in the water. Your new impeller may be toast from running dry.

Were the 2 plastic guide tubes in the water pump housing to make sure copper tubes went into the pump ?

There was only one in place when we disassembled. I looked online but haven’t found them for sale yet

The other one is like up higher on the water tube.----This insignificant part must be in place for proper fit of the water tubes into the pump.
