When doing a compression test should you remove all plugs at one time too do test.

Yes. Remove all plugs. Ground out the leads to the block, or install a spark tester. Screw in the compression tester to 1 cylinder at a time, cranking about 7 to 8 times until the meter reaches its resting spot. Depending on the size and age of the motor, around 100 psi, but mainly you want the cylinders to be within 10 percent of each other.
