ran fine then quit now wont start

Have have a brand new 3.0L with about 4hrs on it. Had not ran it in a week or so went out and put the muffs on it. Had to crank it several times which I figured was just the fuel running back out the the line after sitting a while. Finally got it to start after giving it about half throttle then had a heavy miss.
Started pulling plug wires while it was running and noticed it was'nt hitting on the #1 cylinder very well. Pulled all 4 plugs and did a compression check. All cylinders consistant about 135psi. Tried pouring a little gas in the carb still just runs as long as you crank, although acts like it wants to start. Never got hot or anything. What now??

Do you have spark on all four spark plugs?

Yes but when I pulled the wire on the #1 at idle it did not seem to change the miss where the others did. Then I ran it up to about 2500-3000 rpm and tried it. I could then tell it made a difference when I removed the plug wire.
Everything is brand new except the fuel pump and carb. I forgot to mention I had just reoved the fuel line from the carb to put some teflon on it as it was seeping fuel around the fitting. Maybe air in the fuel line?

DO NOT use Teflon tape on the fuel system. For one thing, there shouldn't be anything there with a pipe thread except the fitting in the fuel pump. Everyting else should be flared tubing.

Try this after you fix your leak.
Remove the valve cover get some rocker arm oil deflector clips to keep oil from going everywhere. Then start the engine and at idle back the #1 lifters off 1 full turn each and see if the idle smooths out or the lifters get noisy.

Yeah I knew you would'nt be impressed with my teflon job as I have read your sticky on fuel system. Why is teflon so bad? The only reason I did that is because I had to put a new fitting on one end and had to cut the double flare off. I could not find anyone with a flare tool good enough to make a flare on that SS line so I used my own Snap On double flare tool (nearly bent the handle in half trying to put the final flare back on it.) Guess it still needs flared wider.
