I don't know to much about glassing or anything, but I would say yes on giving it a rough sand. I know resin seems to stick better to a roughed up surface and everything.

I think its a better idea not to form a filet from PL, its more like a hard putty, too soft for glass to hold onto. Make filets with thickened resin.
When I did mine I wiped all the excess PL away...something tells me sanding it would be difficult, maybe a rasp plane like they use for bondo bodywork will cut it away. Or throw extra glass over it and forgetaboutit.:p

Depends on what kind of access you have to the bead of PL. I bed my stringers in PL, and then use thickened resin or boat bondo to create a fillet for the fiberglass to form over...no sanding required. Trim with a knife if you feel the need to remove excess.

Sanding of any adhesive works poorly...it's an adhesive, period.
