I have broken off the speedometer pick up tube nipple and a section of the tube on my 115 Johnson 2000 year model. I tried unscrewing the nipple, but it broke off so I gently drilled out the remaining nylon nipple. I am planning on glueing or JB Welding the new one in. Will that be OK?

Also, can I buy a piece of tube and splice into the existing tubing that runs to the speedo, or would the restriction of the splice prevent the speedo from registering correctly? I am thinking the splice would be the type used on car vacuum lines where the straight line nipple type splice would be inserted into the two tubes to connect them.

This is not critical. A restriction would just slow down the speedo response to change. You just need a leak free connection.

Just press the new nipple in as it was done at the factory. Using glue or JB weld will just make it harder to remove the next time something happens.
