I removed a Factory Mercury prop 10 pitch, it had the brass locking nut with a stainlees tab that has 2 tabs you bend over brass locking nut and the two slots that fit over the round tits on prop. Replaced with a factory Suzuki prop , that fit right on using Merc rear thrust spacer, "But" now I have no way of locking the nut down to propshaft, and of course there is no hole in propshaft. Is it safe to run with a locking nut only or might it come off??? I have enough threads to add a short second nut on end.

I wouldn't run it without locking the nut in some way, I say add the other nut.
